About the Schlögelhofer lab:

We study mechanisms of somatic and meiotic DNA repair and recombination in the model plant _Arabidopsis thaliana_.

We aim at understanding the mechanistic details by employing biochemical assays with purified proteins to investigate individual steps of DNA break formation and repair.

Recently we have developed single-molecule approaches to study protein-DNA and protein-protein interactions.

Furthermore, we want to understand how individual steps of DSB formation and repair are coordinated, embedded in the overall development (especially during meiosis) and connected to other crucial processes (for instance pathogen defence).

To address these questions we generate (hypo
- and hyper-morphic) mutants and investigate the spatial and temporal distribution of key factors by regular and super-resolution microscopy. We utilise state-of-the-art technology provided by the excellent on-site facilities (next generation sequencing) to investigate DNA repair and recombination genome-wide and to identify new protein factors (mass spectrometry).

About the project:

DNA damage needs to be communicated within a (plant) cell irrespective of the developmental stage.

The conserved DNA damage signalling kinase ATM is the first on-site to provide such a signal following DNA double strand break formation.

While one mode of activation (via the MRE11 complex) appears to be conserved, alternative activation modes have not yet been investigated in plants.

In the course of the MSc project the student will generate crucial research tools to address the question how the ATM kinase can be activated.
