Research Assistant

The **Jachowicz Lab** investigates the regulatory role of “dark” genome elements including transposons, repeats, and other non-canonical RNAs in shaping genome functions during first lineage-specification in mammals. We use a novel, genome-wide technologies based on split-and-pool barcoding that allow simultaneous measurement of 3D nuclear organization and transcription within single cells and combine them with functional assays identifying key lineage specification regulators. We use mouse embryonic stem cells and early mouse embryos as our model systems.

**Research Assistant / Lab Manager (Jachowicz Lab)**:
We are looking for a highly organized person with laboratory experience in molecular/cellular techniques and mouse care. This person will help to run the lab, take care of mouse colony, help with mammalian cell tissue culture, and assist with multiple projects developing cutting-edge multi-omics single cell methods to measure 3D genome structure and transcriptome during early mammalian development.

**Your responsibilities**:

- **Lab maintenance**: help to run the lab by ensuring instruments and equipment are functioning properly, ordering/inventorying reagents and lab supplies, ensuring overall lab compliance with all associated rules and regulations, keeping general lab documentation up to date, mediate interactions with in-house facilities and services
- **Mouse colony management**: breeding, timed breeding, hormonal injections (superovulation), oocyte and preimplantation embryos isolation, collection of samples for genotyping, weaning, aging and sexing of pups, retiring of mouse colonies
- **Routinely handle live mice and early embryos to perform experiments**: embryo isolation, mouth pipetting, immunostaining, fluorescent in situ-hybridization
- **Molecular/cellular techniques**: general knowledge of molecular biology techniques including PCR, RTqPCR, DNA and RNA isolation, WB
- **Basic cell culture**: mammalian cells culture and passaging, media, and reagents reparation preparation
- Possible involvement in specific lab projects focused on development of multi-omics methods to measure 3D DNA-RNA interactions including handling single cell samples, cells crosslinking, split-and-pool barcoding technique, DNA/RNA library preparation

**Your profile**:

- At least 2 years expertise in molecular biology/ cellular biology/ biochemistry/ genomics or other relevant fields
- Will to work with live animals and early mouse embryos
- High level of organization and motivation
- Fluency in English and German

**Additional assets** will be considered a plus such as:

- Interest in technology development
- Interest in early mammalian development and molecular mechanisms
- Experience in mouse handling (early embryo handling is a great plus) or
- Experience in genomics (DNAseq, HiC, RNAseq, ATACseq, CHiPseq or any other
