**Our research**:
Biomolecular condensates are membraneless assemblies that can form via liquid liquid phase separation and have the ability to concentrate biopolymers. Recent studies suggest that condensates play important roles in cellular organization and physiology. However, many aspects of condensate structure, function, assembly and disassembly are poorly understood. A failure to control condensate formation and dissolution may lead to protein misfolding and aggregation. Hence, an improved understanding of condensate pathology could enable the treatment of protein aggregation diseases.
**About the lab**:
The Köhler Lab is particularly interested in chromatin and transcription biology. Recently, we discovered layered condensates, in which chromatin-modifying enzymes encapsulate a liquid scaffold (Gallego et al., Nature 2020). The scaffold attracts chromatin and promotes its modification by ubiquitin. This is an intriguing example of a dynamic nuclear sub-compartment that organizes enzymatic activity and transcription in a unique way. Our current studies focus on the mechanisms that have evolved to regulate the behaviors of nuclear condensates. Specific questions are: how does the cellular protein quality control machinery ensure the integrity of condensates? Why do condensates change their material properties during environmental stress and ageing? How do cells prevent the conversion of condensates into protein aggregates, and what are the differences and commonalities between condensates and aggregates?
The Köhler Lab combines biochemical reconstitution/enzymology, single-molecule biophysical studies, structural biology (cryo-EM and crystallography) and genetic studies as well as high-resolution microscopy to tackle these questions.

The Köhler Lab provides a stimulating environment for successful scientific work. The Max Perutz Labs are located at the Vienna BioCenter Campus which has a 30-year history of scientific excellence and training, and is now Austria’s largest life sciences research center. You will benefit from a range of excellent core facilities. The successful applicant will be a full member of the special research focus on targeted protein degradation. The Max Perutz Labs are home to scientists from over 40 different nations with English as the working language. Vienna was named as city with highest quality of life in the world for many times (Mercer survey). Childcare facilities are available on campus.
The position is endowed with a competitive salary and is available initially for 3 years. It can be turned into a permanent position depending on performance. Lab funding includes an ERC Consolidator Grant.
